O objetivo deste estudo foi avaliar estratégias terapêuticas para o tratamento de infecções broncopulmonares, utilizando a enrofloxacina de ação rápida e sua associação com suporte e fluidoterapia endovenosa ou suporte e solução oral energética e eletrolítica, por meio da mensuração de parâmetros clínicos, hematológicos, bioquímicos e desenvolvimento corporal de neonatos bovinos. Foram utilizadas 35 bezerras da raça Holandesa, monitoradas desde o nascimento até a sexta semana de vida, divididas aleatoriamente nos grupos: grupo CONTROLE; grupo antibiótico; grupo antibiótico + suporte + fluidoterapia endovenosa; grupo antibiótico + suporte + solução oral; e grupo SUPORTE. Os parâmetros zootécnicos foram avaliados do nascimento até a sexta semana de vida, e os parâmetros hematológicos e bioquímicos foram avaliados zero, 24, 72 e 120 horas após diagnóstico da broncopneumonia. Os animais do grupo antibiótico + suporte + solução oral apresentaram menores níveis de eosinófilos e maiores níveis de neutrófilos segmentados em comparação aos animais dos demais grupos. Não houve diferença nos parâmetros zootécnicos avaliados. Neste estudo, o tratamento com antibiótico e solução oral ofereceu aos animais melhor aporte para combater a broncopneumonia, favorecendo o organismo a desenvolver uma resposta imune efetiva diante da infecção.(AU)


The objective of this study was to evaluate therapeutic strategies for treatment of bronchopulmonary infections using fast-acting enrofloxacin and its association with support and endovenous fluid or support and oral energy and electrolytic solution, by measuring clinical, hematological, biochemical and development parameters of bovine neonates. Thirty-five Holstein calves, monitored from birth to six weeks of age, were randomly divided into five groups: control group; antibiotic group; antibiotic group + support + intravenous fluid therapy; antibiotic group + support + oral solution; and support group. The performance parameters were evaluated from birth to the 6th week of age and hematological and biochemical parameters were evaluated 0, 24, 72 and 120 hours after diagnosis of bronchopneumonia. Calves of the antibiotic group + support + oral solution group presented lower levels of eosinophils and higher levels of segmented neutrophils compared to the other groups. There was no difference in performance parameters evaluated. In this study, the treatment with antibiotic and oral solution offered the animals had a better contribution to treat bronchopneumonia, favoring the organism to develop an effective immune response to that infection.(AU)

Mannheimia haemolytica is an important cause of bovine respiratory disease (BRD). BRD is usually a multifactorial disease with host factors and viral infections influencing pathogenesis. Previous studies that have attempted to experimentally induce pneumonia using aerosolized M. haemolytica alone have produced inconsistent results, yet an aerosol model would be useful to study the details of early infection and to investigate the role of innate defences in pathogenesis. The objective of these studies was to develop and characterize an aerosolized M. haemolytica disease model. In an initial study, conventionally raised calves with higher levels of antibody against M. haemolytica leukotoxin developed acute respiratory distress and diffuse alveolar damage, but did not develop bronchopneumonia, following challenge with M. haemolytica serotype 1. Clean-catch colostrum-deprived calves challenged with 1 × 1010 colony forming units of M. haemolytica serotype 1 consistently developed bronchopneumonia, with elevations in rectal temperature, serum haptoglobin, plasma fibrinogen, and blood neutrophils. Mannheimia haemolytica serotype 1 was consistently isolated from the nasal cavities and lungs of challenged calves. Despite distribution of aerosol and isolation of M. haemolytica in all lung lobes, gross lesions were mainly observed in the cranioventral area of lung. Gross and histologic lesions included neutrophilic bronchopneumonia and fibrinous pleuritis, with oat cells (necrotic neutrophils with streaming nuclei), and areas of coagulative necrosis, which are similar to lesions in naturally occurring BRD. Thus, challenge with M. haemolytica serotype 1 and use of clean-catch colostrum-deprived calves with low or absent antibody titres allowed development of an effective aerosol challenge model that induced typical clinical disease and lesions.

RESUMO

Streptococcus suis is a major Gram-positive swine pathogen associated with a wide variety of diseases in pigs. The efforts made to develop vaccines against this pathogen have failed because of lack of common cross-reactive antigens against different serotypes. Nowadays the interest has moved to surface and secreted proteins, as they have the highest chances to raise an effective immune response because they are in direct contact with host cells and are really exposed and accessible to antibodies. In this work, we have performed a comparative immunosecretomic approach to identify a set of immunoreactive secreted proteins common to the most prevalent serotypes of S. suis. Among the 67 proteins identified, three (SSU0020, SSU0934, and SSU0215) were those predicted extracellular proteins most widely found within the studied serotypes. These immunoreactive proteins may be interesting targets for future vaccine development as they could provide possible cross-reactivity among different serotypes of this pathogen.

In the current study, we compared the therapeutic effects of a non-steroidal and a steroidal anti-inflammatory drug on the production of pro-inflammatory cytokines, interleukin-1ß (IL-1ß), interleukin-6 (IL-6), interleukin-12p40 (IL-12p40), interferon gamma (IFNγ), and tumor necrosis factor alpha (TNF-α) in the blood of water buffalo (Bubalus bubalis) calves naturally infected by bronchopneumonia. Twenty-seven buffalo calves (7 ± 2-month-old, 163 ± 12 kg) reared in smallholder farms in El-Dakahlia province in Egypt were identified to have bronchopneumonia and randomly allocated into three equal groups. Ten clinically healthy buffalo calves with negative bronchoalveolar lavage results were served as negative control. Diseased calves were treated with tulathromycin alone, a combination of tulathromycin with dexamethasone (steroidal anti-inflammatory drug) or tulathromycin with flunixin meglumine (non-steroidal anti-inflammatory drug). The results revealed significant elevations (P < 0.05) in the production of selected cytokines in all diseased calves in comparison with healthy animals. Six days post-treatment, a significant inhibition (P < 0.05) in the production of all assessed cytokines was observed in the blood of all treated calves. Interestingly, the serum concentrations of IL-1ß and IL-12p40 were returned to the normal levels in pneumonic calves treated with the combination therapy of tulathromycin and flunixin meglumine. A strong significant positive correlation (P < 0.05) was detected between clinical sum scoring and IL-12p40 and TNF-α concentrations. The obtained results indicate the selectively potent anti-inflammatory effect of flunixin meglumine on the production of pro-inflammatory cytokines in pneumonic buffalo calves and highlight the efficacy of flunixin meglumine in the treatment of bronchopneumonia in buffalo calves when used in combination with tulathromycin.

A retrospective study was performed to compare the treatment regimens in feedlot cattle that died with bovine respiratory disease (BRD) to the antimicrobial susceptibility patterns of the microorganisms isolated from lungs. Forty-three cattle submitted by the Willard Sparks Beef Research Center (WSBRC) to the Oklahoma Animal Disease Diagnostic Laboratory for postmortem examination during 2007 had bronchopneumonia (acute = 16, subacute = 5, or chronic = 22). Lungs from cattle were cultured aerobically (40 cattle) and for Mycoplasma spp. (34 cattle). Susceptibility panels were performed. At least 1 BRD pathogen (Mannheimia haemolytica, Pasteurella multocida, Histophilus somni, Mycoplasma bovis, or Arcanobacterium pyogenes) was isolated from 39 cattle, and 77% (30/39) had multiple organisms recovered. Mycoplasmal infections were common (25/34) and a major component of mixed infections (24/25). The majority (60%) of the M. haemolytica, P. multocida, and H. somni isolates were resistant to tetracycline. Most of the H. somni isolates (67%) were susceptible to tilmicosin (Ti), enrofloxacin (En), ceftiofur (Ce), and florfenicol, despite extensive treatment with Ti, En, and Ce (75% of isolates were from cattle that received each antimicrobial once). Most of the M. haemolytica (65%) and P. multocida (79%) isolates were susceptible to En and Ce, despite antemortem treatment of cattle with these antimicrobials. Hence, the current study reports a discrepancy between the antemortem treatment of clinical BRD and the susceptibility patterns of the bacteria isolated from lungs postmortem. Based on these findings, factors other than antimicrobial resistance are playing a role in the death of feedlot cattle with BRD.

Estudou-se a broncopneumonia de ocorrência natural em bezerros e a influência da administração de acetato de DL-α-tocoferol na profilaxia e tratamento desta enfermidade. Foram avaliados 60 bezerros machos, com idade máxima de 10 dias, divididos em dois grupos experimentais: GSV (grupo sem vitamina) e GCV (grupo com vitamina). A suplementação com dose única de 4.500 UI de acetato de DL-α-tocoferol por via intramuscular (IM) foi precedida por exame físico, perfil hematológico, determinação de proteínas séricas, globulinas, gamaglutamiltransferase e citologia do lavado traqueobrônquico (D0) para verificar a sanidade e homogeinização dos grupos. Os bezerros foram mantidos em bezerreiro e distribuídos aleatoriamente, 2 a 2 em baias individuais de 2,40m², onde permaneceram até o 21º dia (D0 ao D21). Foram avaliados por exame físico diário e, na presença de sinais clínicos indicativos de broncopneumonia (DX), retirados do bezerreiro, avaliados por hemograma e citologia do lavado traqueobrônquico, e tratados com enrofloxacina (5mg/kg, IM). Durante o tratamento, o exame físico foi realizado diariamente, e o hemograma e a citologia do lavado traqueobrônquico repetidos uma semana após seu término (DY). Nos bezerros sadios foram repetidos hemograma e citologia do lavado traqueobrônquico, no último dia (D21) do experimento. Não houve diferença significativa quanto às variáveis gamaglutamiltrasfrase (p=0,09), proteínas séricas (p=0,27) e globulinas (p=0,10) e a idade dos bezerros (p=0,15) entre os grupos. Os bezerros do grupo GSV e GCV levaram em média 11 e 12 dias para adoecerem, respectivamente. Contudo, não houve diferença estatística significativa entre os grupos (p=0,68). Em 34 bezerros (56,66 por cento) foi diagnosticado broncopneumonia, sendo que 17 destes bezerros (50 por cento) pertenciam ao grupo GCV e 17 (50 por cento) ao grupo GSV. Com relação aos sinais clínicos, não houve diferença significativa entre os grupos em nenhum dos momentos estudados. Dos 34 bezerros que adoeceram 73,52 por cento morreram pela doença, sendo 64,7 por cento do GCV e 82,35 por cento do GSV. A citologia do lavado traqueobrônquico não apresentou diferença significativa entre os grupos em nenhum dos momentos estudados (D0, DX, DY e D21). A suplementação com vitamina E não teve influência na profilaxia e no tratamento de broncopneumonia de bezerros.


The natural occurrence of bronchopneumonia in calves and the effect of DL-α-tocopherol in the prophylaxis and treatment of this disease is studied. We evaluated 60 male calves, aged up to 10 days, divided into two groups: GSV (group without vitamin) and GCV (group with vitamin). Supplementation with a single dose of 4,500 IU of DL-α-tocopherol administered intramuscularly (IM) was preceded by clinical examination, complete blood count (CBC), determination of serum proteins, globulins, gamaglutamyltransferase and tracheobronchial lavage cytology (D0) to check sanity and homogenization of the groups. The animals were kept two by two in individual 2.40 m² stalls, where they remained until day 21 (D0 to D21). The animals were examined daily and, in the presence of clinical signs of bronchopneumonia (DX), taken from stalls, evaluated by CBC and tracheobronchial lavage cytology, and treated with enrofloxacin (5 mg/kg, IM). During the treatment, clinical examination was performed daily by the CBC and tracheobronchial lavage cytology repeated one week after the end of treatment (DY). In healthy animals, CBC and tracheobronchial lavage cytology were repeated on the last day of the experiment (D21). No significant difference for the blood variables gamaglutamiltrasfrase (p=0.09), serum protein (p=0.27) and globulin (p=0.10) and the age of animals (p=0.15) were observed in both of groups. Animals in group GSV and GCV took an average of 11 and 12 days for get sick, respectively. However, no statistical difference between was observed in both groups (p=0.68). In 34 (56.66 percent) animals were diagnosed bronchopneumonia, and 17 of these calves (50 percent) belonged to group GCV and 17 (50 percent) to group GSV. With respect to clinical signs, there was no significant difference between groups in any of the times studied. Out of 34 animals which were affected, 73.52 percent died of the disease, 64.7 percent were from GCV group and 82.35 percent were from GSV group. Cytology of tracheobronchial lavage showed no difference between groups in any of the times studied (D0, DX, DY and D21). We conclude that supplementation with vitamin E had no influence in the prophylaxis and Treatment of bronchopneumonia in calves.

RESUMO

This case report describes gross lesions and histopathological findings in a 3-months-old calf originating from a feedlot with approximately 400 cattle. In this animal and additional 14 cattle of similar age, which were kept together in the same stable, swollen joints had occurred suddenly. The examination of this calf showed that a severe polyarthritis induced by haematogenous spread of Mycoplasma bovis following bronchopenumonia was present, which was characterised by necrotising lesions of the joint capsules and severe cartilage erosions.

Mycoplasma bovis is known to be responsible for pneumonia and arthritis in calves, as well as mastitis in dairy cows. Despite clear evidence of its pathogenic potential, little is known about mechanisms of cytadherence and the molecular factors involved. The purpose of this work was to compare adherence rates of M. bovis field strains to different host cell lines and study the effects of cloning and sub-culturing M. bovis strains on their adherence properties. Eighteen metabolically labeled M. bovis strains isolated from different pathological backgrounds were examined in adherence trials using four different host cell lines, i.e. embryonic bovine lung (EBL), embryonic bovine trachea (EBTr), Madin Darby bovine kidney (MDBK) and rabbit kidney (RK) cells. Although large interstrain variations in adherence rates (3.4-19.1%) were measured they could not be correlated to the pathological background (pneumonia, arthritis or mastitis). Adherence rates to the fibroblast cell line (EBTr) were significantly lower than those to the three epithelial cell lines (EBL, MDBK and RK). The only non-pathogenic strain (221/89) exhibited lower adherence rates than three isolates from clinical mastitis. Interestingly, adherence rates were significantly reduced after in vitro passaging. In contrast, no effect of single cloning of strains on adherence was observed. There was no general correlation between expression of variable surface proteins (Vsps) as monitored by immunoblotting and adherence rates, although alterations in Vsp expression profiles were seen as a consequence of passaging. As there is probably a large number of adhesins, variable and non-variable, on the surface of M. bovis cells the issue is very complex, and the most active components have yet to be identified.

A study was made of the influence of UV-irradiation (254 nm) of blood in vitro, of the autotransfusion of UV-irradiated blood (AUVIB), and of the mixture of UV-irradiated and intact blood in vitro on the content of bactericidal cation proteins (CP) in blood neutrophil of calves suffered from dyspepsia and broncho-pneumonia. Age differences were noticed in CP contents and their decrease in neutrophils following AUVIB in vivo and administration of the mixture of blood in vitro. The decrease in cell CP contents is presumably due to neutrophil degranulation and CP release into the blood plasma. Since the initial mechanisms of neutrophil degranulation are located on the cell surface, the CP release is supposed to result from a membranotropic effect of UV-irradiated blood on the intact autologous blood. This effect may explain the increase in nonspecific resistance of organism after the AUVIB, being one of the main therapeutic phenomena of the AUVIB-therapy.

In a diagnostic survey of diseases in wild white-tailed deer (62 cases) and mule deer (12 cases) the most common findings were traumatic injury (20%), nontraumatic hemorrhage (13%), polioencephalomalacia (11%), and bacterial infections (9%). Although epizootic hemorrhagic disease was suspected in several cases, the virus was isolated from only 1 white-tailed deer.

Studied was the effect of lactoplasmin, gamma-globulin, a tissue preparation of swine embryos after Filatov, and a hyperimmune rabbit serum on the hemopoieis, the white blood picture, and the morbidity rate in calves with bronchopneumonia. It was found that these preparations influence favourably the white blood picture, the clotting time of blood, and the resistance of calves. With the test calves the morbidity rate was lower than in the controls. Best prophylactic effect was produced with use of tissue preparations and lactoplasmin.
